If it's a standard coil/wick setup I try to rewick. Usually reuse the coil unless it's all gunked up and nasty. What kind of tank and coil setup? If it's a prebuilt coil/wick like used in clearomizers or other disposables it may or may not be able to be rewicked. If you just add different juice to a wick that was previously used for a different flavor the old flavor will still come through and it's not always pretty. Usually when switching flavors I try to switch the wick at least or replace the wick/coil if I can't rewick it.
Worst case scenario if you have no other coils/wicks and you're out of the old flavor and no choice but to switch flavor, you 'could' just add the new juice. Technically it should vape, how good it tastes on the other hand is a roll of the dice.

Be careful when you pull the wick out if it's not real old as you can unwind the coil. I hold the coil with my thumbnail and pull it out. If it's stubborn I cut it off as close to the coil as I can on one side and then pull it out the other. Then I heat it up and put it under hot water and use an old toothbrush to clean it off. Then hear it up just a bit to get water off of it. Then rewick. Start with new juice.
Prebuilt coil I generally wash in alcohol and hot water. I do rebuild some Prebuilt coils. But for the most part I'm lazy and caring a spare is easier.

Unwind the coil? How so? I've had some smaller factory made coils that have broken or bunched up to one side (horizontal coils) from tugging the wick out a bit too hard. Since I've been able to build my coils on a real deck my wick hasn't stuck yet. Maybe because I'm wicking it a bit looser or because I like my coils with a big hole. Then again some builds require longer tufts of cotton to be wicked and tucked down, my deck uses short amounts of wick so maybe that's why it doesn't get stuck. It's almost cut off near the end of the coil to start with.
